Lack of association between ACE insertion/deletion polymorphism and lung cancer: A meta-analysis.

Previous case-control studies on the relation between the angiotensin-converting enzyme (ACE) gene insertion/deletion (I/D) polymorphism and lung cancer were controversial. In this study, we aimed to further evaluate the relation between the ACE gene I/D polymorphism and lung cancer. We selected eight case-control studies related to the ACE gene I/D polymorphism and lung cancer by searching PubMed, EMBase, Chinese Biomedical Literature Database, and Wanfang database. We utilized Q-test and I (2) test to test the heterogeneity between each study. To merge the odds ratio (OR) and 95% confidence interval, we utilized the fixed-effects model during the analyses. The present study included 1612 patients with lung cancer and 1442 cancer-free control subjects. By meta-analysis, we did not find any association between the ACE gene I/D polymorphism and lung cancer in either genotype or allele distribution (DD vs. (ID+II): OR = 0.96, 95% CI (0.80-1.14), p = 0. 61; D allele vs. I allele: OR = 1.01, 95% CI (0.91-1.13), p = 0.79). We concluded that the ACE gene I/D polymorphism was not associated with lung cancer.

Lack of association of angiotensin-converting enzyme insertion/deletion polymorphism with breast cancer: An update meta-analysis based on 10405 subjects.

Previous case-control studies on the relation between angiotensin-converting enzyme (ACE) gene insertion/deletion (I/D) polymorphism and breast cancer did not reach the same conclusion. In the present study, we aimed to further evaluate the relationship between the ACE gene I/D polymorphism and breast cancer. We selected 13 case-control studies related to ACE gene I/D polymorphism and breast cancer by searching PubMed, EMBase, Chinese Biomedical Literature Database, Chinese CNKI, and Wanfang database. To test the heterogeneity between each study, we utilized the Q-test and I(2) test. To merge the odds ratio (OR) and 95% confidence interval (CI), we utilized the random effects model during the analyses. The present study included 1997 patients with breast cancer and 8404 cancer-free control subjects. By meta-analysis, we did not find any association of ACE gene I/D polymorphism with breast cancer in different model (DD vs (ID+II): OR=1.28, 95% CI (0.90-1.81), p=0.16; II vs (ID+DD): OR=0.99, 95% CI (0.81-1.21), p=0.93; D allele vs I allele: OR=1.15, 95% CI (0.94-1.41), p=0.18). We concluded that ACE gene I/D polymorphism was not associated with breast cancer.

Association of ACE insertion or deletion polymorphisms with the risk of coronary restenosis after percutaneous coronary intervention: A meta-analysis.

Previous case-control studies on the relationship between the angiotensin-converting enzyme (ACE) gene insertion/deletion (I/D) polymorphisms and coronary restenosis did not reach the same conclusion. In the present study, we aimed to further evaluate the relationship between the ACE gene I/D polymorphisms and coronary restenosis, after percutaneous coronary intervention (PCI). By searching PubMed, EMBase, the Chinese Biomedical Literature Database and Wanfang database, we selected 16 case-control studies related to ACE gene I/D polymorphism and coronary restenosis after PCI. To test for heterogeneity in each study, we utilized the Q-test and I(2) test. To merge the odds ratio (OR) and 95% CI, we utilized the random effects model during the analyses. The present study included 4693 subjects: 1241 patients with coronary restenosis and 3452 without coronary restenosis. By meta-analysis, we found there was significant association of ACE gene I/D polymorphism with coronary restenosis (D allele versus I allele: OR = 1.92; 95% CI (1.40-2.43); p < 0.001). A subgroup analysis, by stratification according to ethnicity, also showed that this association was found not only in the Caucasian population ((D allele versus I allele: OR = 1.94; 95% CI (1.38-2.80); p < 0.001)), but also in the Asian population ((D allele versus I allele: OR = 1.83; 95% CI (1.05-3.20); p = 0.03)). After stratification according to age, we found that the D allele carriers have a higher risk for development of coronary restenosis in subjects < 60 years old (OR = 2.13; 95% CI: 1.40-3.24; p = 0.0004); while in the subjects ⩾ 60 years old, the association was present with bordering significance (OR = 1.48; 95%CI: 0.98-2.25; p = 0.06). The present study suggested that the ACE gene I/D polymorphism was associated with coronary restenosis, regardless of age and ethnicity.

Angiotensin-converting enzyme insertion/deletion polymorphism and risk of myocardial infarction in an updated meta-analysis based on 34993 participants.

The association between angiotensin-converting enzyme insertion/deletion (ACE I/D) polymorphism and risk of myocardial infarction (MI) has been extensively studied. However, the results were in controversy. This study aimed to explore the association between ACE I/D polymorphism and risk of MI by using a meta-analysis. We retrieved the following databases to indentify eligible studies: Medline, Embase, ISI, VIP, CBM and Wan Fang database. The latest update was 10th May, 2012. Odds ratio and 95% confidence interval (95% CI) were used to present the strength of the association. A total of 40 case-control studies with 34993 participants were included. Overall, D allele of ACE I/D polymorphism was significantly associated with an increased risk of MI in genetic comparison models (OR (95% CI): 1.41 (1.22-1.64) for DD vs. II; 1.11 (1.01-1.21) for ID vs. II; 1.23 (1.10-1.37) for D carriers vs. II; 1.28 (1.15-1.43) for DD vs. I carriers and 1.06 (1.02-1.10) for D carriers vs. I carriers). Subgroup analyses, according to ethnicities and countries of participants also indicated that D allele was significantly associated with an increased risk of MI in Asians (especially for Chinese) and Caucasians (especially for English, French, Germans and Italians) (OR (95% CI) of DD vs. ID+II: 2.11 (1.65-2.70) for Asians and 1.15 (1.05-1.27) for Caucasians). In conclusion, this meta-analysis indicated that D allele of ACE I/D polymorphism was a possible risk factor for MI incidence for both Asians and Caucasians.
