Variations of melanocortin receptor 1 (MC1R) gene in three pig breeds.

Variations of Melanocortin Receptor 1 (MC1R) were investigated using sequencing, PCR-RFLP and PCR-SSCP, in three pig breeds, Landrace, Yorkshire, and Duroc. Five polymorphic sites were found, in which 668G-->C occurred within 5' UTR, nt894insCC in coding region resulting in a premature stop at codon 56, and 1318C-->T, 1554G-->A, 1197G-->A in coding region resulting in Ala164Val, Ala243Thr, and Asp124Asn respectively. All individuals in Landrace and Yorkshire present homozygous 668GG, 1197AA, 1318CC, and 1554GG, and have CC insertions at the 894 site, whereas the individuals in Duroc present a contrast homozygous 668CC, 1197GG, 1318TT, and 1554AA, and have no CC insertions at the corresponding site. No heterozygote has been found at these mutation sites. Presumably, 668G-->C, 1318C-->T, and 1554G-->A may be associated with the recessive red color in the Duroc breed, and nt894insCC making 1197G-->A nonsense may be associated with the white color in Landrace and Yorkshire breeds.

[Studies of the relationship of melanocortin receptor 1(MC1R) gene with coat color phenotype in pigs].

Although coat color in pigs has no direct relation with economic traits, it affects economic benefit significantly, coat color selection are widely used in pig breeding and production. PCR-Acc II-RFLP, PCR-BspH I-RFLP and PCR-SSCP were used in combination to analyze genotype at MC1R locus among individuals from 16 full-sib pedigrees and 6 Chinese native breeds including Jinhua, Jiaxing Black, Yushan Black, Leping Spotted, Shanggao Spotted and Shengxian Spotted pig. It was found that the Chinese native pig breeds carry a dominant black allele at MC1R at high frequency, this ED1 allele was suggested to be the major allele controlling black coat color in Chinese native pig breed. In addition, the evidence for a new allele was obtained in Shengxian Spotted pigs by PCR-SSCP analysis. It was reconfirmed from the result of pedigree analysis that ED1 was dominant over EP and e, while EP was incompletely dominant over e.

Genetic variation within coat color genes of MC1R and ASIP in Chinese brownish red Tibetan pigs.

Melanocortin receptor 1 (MC1R) and agouti signaling protein (ASIP) are two major genes affecting coat color phenotypes in mammals, and inactivation mutations in the MC1R gene are responsible for red coat color in European pig breeds. Conversely, the gain-of-function ASIP mutations block MC1R signaling and lead to the production of red pheomelanin. Chinese Tibetan pigs have three types of coat color phenotypes, including brownish red, solid black and black with patches of brownish red and white. Herein, we investigated variations of the MC1R and ASIP genes in Tibetan pigs. The results showed that the brownish red Tibet pig had the dominant black MC1R allele (E(D1)). No loss-of-function mutation in MC1R responsible for red coat color in European breeds was observed in this breed. No causal mutation for the red coat color phenotype was found in the coding sequence of the ASIP gene. A novel missense mutation c.157G > A was firstly identified in exon 2 of ASIP, which was further genotyped in 285 pigs from five Chinese breeds and three Western breeds having different coat color phenotypes. Nearly all pigs were GG homozygotes. In conclusion, no functional variant responsible for brownish red coloration was found in the coding region of MC1R and ASIP in Tibetan pigs.

[Effect study of white locus (I) on coat color inheritance in Chinese native pig breeds].

The classical White (I) locus is one of the important pig coat color hereditable loci,which is homologous to KIT gene. In this study,PCR-RFLP and PCR-SSCP analysis were commanded on the Intron 17 and 18 nucleotide sequences of KIT gene. The tested results showed that the substitution mutation (G-->A) of Intron 17 was found in white pigs, including Wuzhishan pigs (white), Landrace and Large White,and its genotype (AB) frequency was 1.1 and 0.8 respectively. The genotype frequency was uniformly 0 in the other native pig breeds. Similarly,the deleted mutation (AGTT) of Intron 18 was also found in the same white pigs,its genotype (,AA) frequency was 1.1 and 0.93 separately,while in the other native pig breeds was 0. Accordingly it was considered that KIT gene was an important factor regulating the white coat color genotype, and the classical I locus (KIT gene) was epistatic to the other genetic coat color loci. On the other hand,although the native pig breed Rongchang Pig is similar to Landrace and Large White in phenotype (white coat color), the mutation status found in them was absolutely distinct. So it is presumed that the coat color genetic system of Chinese native pig breeds was different from that of imported breeds.

[Research on genetic variation of heart fatty acid-binding protein gene in ten pig breeds].

The genetic variation of heart fatty acid-binding protein (H-FABP) gene in 561 pigs including Duroc, Landrace, Large Yorkshire, Nanchang white pig, Erhualian, Meishan, Yushan black pig, Leping spotted pig, Jinhua black head-hind pig and Shanggao black head-hind pig were detected by PCR-RFLP with Hinf I, Hae III and Msp I. The results showed as follows: (1) Nanchang white pig presented only HH genotype while other breeds varied at the Hinf I-RFLP site; (2) The exotic breeds including Duroc, Landrace, Large Yorkshire and the native breed Nanchang white pig were proved to be polymorphic while the five Chinese local breeds presented no variation at the Hae III-RFLP site; (3) Among the tested breeds only Duroc presented variation at the Msp I-RFLP site. It is noted that all the Chinese local breeds present as AADD--genotypes.
