Fertility following fixed-time AI or insemination at observed estrus in Ovsynch and Heatsynch programs in lactating dairy cows.
The objective of this study was to compare the conception rate for fixed-timed artificial insemination (FTAI) and observed heat artificial insemination (HAI) prior to the scheduled FTAI in Ovsynch and Heatsynch synchronization protocols. In Experiment 1, lactating dairy cows (n=535) received two set-up injections of 25mg prostaglandin F(2alpha) (PGF(2alpha)) i.m., 14 days apart starting at 36+/-3 days in milk (DIM). Cows were blocked by parity and were randomly allocated to either Ovsynch or Heatsynch groups. All cows received 100 microg of GnRH i.m. 14 days after the second set-up injection of PGF(2alpha), followed by a third injection of 25mg PGF(2alpha) i.m., 7 days later. In the Ovsynch group, HAI cows (n=29) were bred on standing estrus after the third PGF(2alpha) before the scheduled second GnRH, whereas FTAI cows (n=218) that were not observed in estrus, received a second injection of 100 microg of GnRH i.m., 48 h after the third PGF(2alpha) and received TAI 8 h after the second GnRH. In the Heatsynch group, all cows (n=288) received 0.5 mg of estradiol cypionate (ECP) 24 h after third PGF(2alpha) and HAI cows (n=172) were bred on standing estrus and FTAI cows (n=116) that were not observed in estrus, received TAI 72 h after the third PGF(2alpha). In Experiment 2, repeat breeder cows (n=186) were randomly assigned to either Ovsynch or Heatsynch groups. The FTAI and HAI cows were inseminated similar to Experiment 1. All cows were observed for estrus three times daily. The associations with the conception rate were modeled with logistic regression separately for Experiments 1 and 2. Of all the variables included in the model in Experiment 1, type of AI (HAI versus FTAI, P=0.0003) and parity (primiparous versus multiparous, P=0.05) influenced the first service conception rate. Over-all conception rate and first service conception rate for HAI cows were higher compared to FTAI cows (33.8% versus 21.3%, and 35.3% versus 21.0%; P=0.001). In the Heatsynch group, cows that received HAI had significantly higher over-all conception rate and first service conception rate compared to FTAI (35.2% versus 17.3% and 36.0% versus 15.5%; P=0.0001). The conception rates in repeat breeder cows for HAI and FTAI (30.1% versus 22.3%) were not different (P>0.1). In conclusion, it was recommended to include AI at observed estrus and fixed-time AI for cows not observed in estrus in order to improve the conception rate in synchronization protocols.

Inseminations at estrus induced by presynchronization before application of synchronized estrus and ovulation.
A controlled field study examined conception rates after 2 timed artificial insemination (TAI) breeding protocols conducted on 2 commercial dairy farms. Estrous cycles in postpartum lactating cows were presynchronized with 2 injections of PGF(2alpha) given 14 d apart (Pre-synch) and then, after 12 d, the standard Ovsynch protocol (injection of GnRH 7 d before and 48 h after an injection of PGF(2alpha), with one TAI at 12 to 16 h after the second GnRH injection) or Heatsynch protocol [injection of GnRH 7 d before an injection of PGF(2alpha), followed 24 h later by 1 mg of estradiol cypionate (ECP) and one TAI 48 h after ECP] was applied. Experimental design allowed artificial insemination to occur anytime after the second Presynch injection and during the designed breeding week when estrus was detected. Of the 1846 first services performed, only 1503 (rate of compliance = 81.4%) were performed according to protocol. Numbers of cows inseminated, logistic-regression adjusted conception rates, and days in milk (DIM) were for inseminations made: 1) during 14 d after first Presynch injection (n = 145; 22.6%; 54 +/- 0.4 DIM); 2) during 12 d after second Presynch injection (n = 727; 33%; 59 +/- 0.2 DIM); 3) during 7 d after the first GnRH injection of Ovsynch or Heatsynch (n = 96; 32.1%; 74 +/- 0.5 DIM); 4) after estrus as part of Heatsynch (n = 212; 44.6%; 76 +/- 0.3 DIM); 4) after TAI as part of Heatsynch (n = 154; 21.1%; 76 +/- 0.4 DIM); 5) after estrus as part of Ovsynch (n = 43; 48.7%; 77 +/- 0.7 DIM); and 6) after TAI as part of Ovsynch (n = 271; 24.4%; 77 +/- 0.3 DIM). Conception rates when AI occurred after one Presynch injection were less than when AI occurred after 2 Presynch injections. Conception rates for those inseminated after either Presynch injection did not differ from those inseminated after combined Heatsynch + Ovsynch. Cows in the Ovsynch and Heatsynch protocols inseminated after estrus during the breeding week had greater conception rates than those receiving the TAI, but overall conception rates did not differ between protocols. Among cows inseminated after detected estrus, conception was greater for cows in the Heatsynch + Ovsynch protocol (77 +/- 0.4 DIM) than for those inseminated after either Presynch injection (54 +/- 0.4 or 59 +/- 0.2 DIM). We concluded that conception rates after Heatsynch and Ovsynch were similar under these experimental conditions, and that delaying first AI improved fertility for cows inseminated after detected estrus.

Effect of presence of clinical and subclinical endometritis at the initiation of Presynch-Ovsynch program on the first service pregnancy in dairy cows.
The present study examined the effect of presence of clinical or subclinical endometritis at the initiation of Presynch-Ovsynch estrous synchronization program on the first service pregnancy rate in dairy cows. Lactating Holstein cows (N=275) were given a thorough reproductive examination at 32-38 days in milk, 3 days prior to the scheduled start of Presynch-Ovsynch program. Based on the reproductive exam findings the cows were diagnosed and classified into three groups as clinical endometritis, subclinical endometritis and normal. All cows received two set-up injections of 25mg PGF(2alpha) (Lutalyse((R)), Pfizer Animal Health, New York, NY, USA) i.m., 14 days apart starting at 35-42 days in milk (DIM). All cows received 75microg of GnRH (Cystorelin, Merial, Iselin, NJ, USA) i.m. 14 days after the second pre-synchronization injection of PGF(2alpha), followed by a third injection of 25mg PGF(2alpha) i.m. 7 days later. Cows received a second injection of 75microg of GnRH i.m. 54h after the third PGF(2alpha), and received timed artificial insemination at the time of the second GnRH injection or 24h later. Multivariate logistic regression was used to analyze the odds of pregnancy at the first service. Variables included in the model were endometritis status (clinical endometritis, subclinical endometritis and normal), farm (two), presence of corpus luteum (CL, yes or no), timing of second GnRH in relation to AI (0 or 24h), sire fertility (bulls with greater compared with lesser estimated relative conception rates), parity (primiparous and multiparous) and their interactions. Of all variable included in the model, cows with corpus letuem (OR=1.83 versus OR=1.00; P=0.05) 3 days prior to the scheduled start of Presynch-Ovsynch program and primiparous cows (OR=1.00 versus OR=0.55; P=0.04) had increased odds of becoming pregnant at the first service. No differences were found in the odds of first service pregnancy among clinical, subclinical endometritis and normal cows (P>0.1). In summary, presence of clinical or subclinical endometritis at the initiation of Presynch-Ovsynch estrous synchronization program does not harm the first service pregnancy rate in dairy cows.

Timed artificial insemination with estradiol cypionate or insemination at estrus in high-producing dairy cows.
A total of 799 Holstein cows from 3 herds were randomly assigned at 37 +/- 3 d in milk (DIM) to timed artificial insemination (AI) or insemination at detected estrus. Cows were presynchronized with injections of PGF(2alpha) at 37 and 51 DIM. At 65 DIM, cows received an injection of GnRH, followed 7 d later by PGF(2alpha). Cows in the estrus-detected group were inseminated after being observed in estrus during the 7 d after the last PGF(2alpha). Cows in the timed AI group received an injection of 1 mg of estradiol cypionate (ECP) 24 h after the last PGF(2alpha). If detected in estrus <or= 24 h after ECP, cows were inseminated then or at a fixed time 48 h after ECP. Pregnancy was diagnosed by ultrasonography at 30 d and by palpation at 44 and 58 d after AI. Plasma progesterone was measured in 4 samples per cow collected on days of 1) second PGF(2alpha), 2) GnRH, 3) third PGF(2alpha), and 4) 48 h after third PGF(2alpha). Cows were classified as cyclic or anovulatory based on progesterone concentrations in samples 1 and 2. Similarly, cows were classified according to progesterone concentrations in samples 2, 3, and 4 (H = >or=1 ng/mL; L = <1 ng/mL), resulting in 8 combinations (LLL, LHL, LLH, LHH, HHH, HHL, HLH, and HLL). Conception rates and pregnancy rates were higher for cows in the timed AI group than in the estrus-detected group at 30, 44, and 58 d (e.g., at 58 d, pregnancy rates were 42.2% for multiparous cows or 34.4% for primiparous cows in the group receiving ECP and timed AI compared with only 20.8 or 18.8% for respective parity subgroups for the treatment group inseminated only at detected estrus). Pregnancy losses were 11.5% from 30 to 58 d and did not differ between treatments. Cyclic cows within both treatments had higher estrous responses, conception rates, and pregnancy rates. Cows that responded to presynchronization and to luteolysis (HHL) had the highest conception and pregnancy rates, followed by cows classified as LHL. Use of 1 mg of ECP to induce ovulation as part of a synchrony regimen improved reproduction at first postpartum insemination in dairy cows.

Effect of gonadotropin-releasing hormone or prostaglandin F(2α)-based estrus synchronization programs for first or subsequent artificial insemination in lactating dairy cows.
The objective was to evaluate the effects of GnRH or PGF(2α)-based synchronization and resynchronization programs on fertility in lactating dairy cows. For experiment 1, cows (n=1,521) were presynchronized with 2 injections of PGF given at 36 and 50 DIM and assigned to 1 of 3 protocols: Ovsynch [OVS; n=552; GnRH injection, PGF(2α) injection 7 d later, GnRH injection 56 h later, and timed artificial insemination (TAI) 16 h later] beginning at 14 d after presynchronization (PS), GnRH-GnRH-PGF(2α)-GnRH (GGPG; n=402) treatment with a GnRH injection given 7 d after PS and OVS 7 d later, or PGF(2α)-GnRH-PGF(2α)-GnRH (P7GPG; n=567) treatment with a PGF(2α) injection given 7 d after PS and OVS beginning 7 d later. Experiment 2 cows (n=2,327) were assigned to 1 of 3 resynchronization protocols 7 d before nonpregnancy diagnosis (NPD): GGPG cows (n=458) received a GnRH injection at enrollment and OVS at NPD; P7GPG cows (n=940) received a PGF(2α) injection at NPD and OVS 7 d later; and P11GPG cows (n=929) received a PGF(2α) injection 3 d after NPD and OVS 11 d later. In both experiments, cows were artificially inseminated upon estrus detection (ED). In experiment 1, 52.3% of cows were artificially inseminated upon ED, with GGPG having reduced ED (GGPG=46.8 vs. OVS=50.7 and P7GPG=57.7%). Treatments did not affect overall pregnancy per artificial insemination (P/AI) at 36 and 66 d after AI (OVS=34.1 and 32.3, P7GPG=34.6 and 31.9, and GGPG=31.3 and 28.1%, respectively) or pregnancy loss but cows artificially inseminated upon ED had higher P/AI than cows undergoing TAI (ED cows=37.9 vs. TAI cows=28.8%). Treatment did not affect P/AI for cows artificially inseminated upon ED or TAI at 36 and 66 d after AI (OVS=34.1 and 32.3, P7GPG=34.6 and 31.9, and GGPG=31.3 and 28.1%). Median days in milk at first AI was affected by treatment (P7GPG=59 vs. OVS=68 and GGPG=68 d). In experiment 2, GGPG reduced ED (GGPG=23.3 vs. P7GPG=74.9 and P11GPG=79.6%). Treatment did not affect overall P/AI at 36 and 66 d after AI (GGPG=29.2 and 25.8, P7GPG=28.7 and 26.6, and P11GPG=31.9 and 30.2%) or pregnancy loss. Cows artificially inseminated upon ED had greater P/AI than TAI cows (ED=32.3 and TAI=25.1%). However, treatment did not affect P/AI for cows artificially inseminated upon ED at 36 and 66 d after AI (GGPG=29.6 and 27.3, P7GPG=29.4 and 28.1, and P11GPG=35.7 and 33.7%) or TAI (GGPG=29.1 and 25.3, P7GPG=25.0 and 22.1, and P11GPG=16.9 and 16.9%). Median days between NPD and AI was affected by treatment (GGPG=10 vs. P7GPG=4 and P11GPG=7 d). Prostaglandin-based programs increased ED and reduced interval to first AI and between AI. Gonadotropin-releasing hormone-based programs increased the proportion of TAI cows. Cows artificially inseminated upon ED had increased P/AI compared with TAI cows.
